Core Facility Services and Fees

Big Dye Terminator V3.1

Users can purchase Big Dye from the Core facility for $85/100 ul or $650/800 ul, or they can purchase >10 (800 ul) vials for $630/vial. There is Big Dye buffer available at no extra cost if you purchase Big Dye from the Core Lab.

 

96-well Sequencing Plates

ABI 96-well sequencing plates are available for purchase in the DNA Core lab for $5.50/plate. Sometimes we have sale plates that are only $3.50/plate (these are purchased from ABI in bulk when they have special promotions).

 

Sequencing options

1) Samples Ready for Sequencing: The Core facility receives purified cycle-sequenced products and provides .ab1 sequence files; optional files available on request are FASTA, Standard Chromatogram (.scf) or Phred (.phd.1). Users complete sample preparation and the Core Lab tech loads them on the machine; users also enter sample names into the computer if they want sample names along with well ID numbers which are automatic. Ask a Core LabTech for a sample sheet template if you want to enter sample names on your own computer ahead of time. HiDi Formamide is provided by the Core Lab at no extra cost. 2) Clean PCR or Plasmid samples for Cycle Sequencing, Sephadex clean-up, and Sequencing: The Core facility receives purified PCR product and primers at specified concentrations with a gel image for reference and then provides .ab1 sequence files; other optional files available on request FASTA, Standard Chromatogram (.scf) or Phred (.phd.1). In this case the Core Lab completes all cycle sequencing, purification, sample preparation and loading. Cost is $10.00 per sample (each sample and primer sequenced) in a tube, or $8.00 per sample in a 96-well plate (minimum 48 samples). Fragment Analysis ( Microsatellites, AFLPs, T-RFLPs)

The user provides premixed samples (PCR product , size standard and HiDi Formamide) in a 96-well plate. The Core Lab tech then loads and runs the samples. All PCR supplies and size standards are provided by the user. HiDi Formamide is provided by the Core Lab at no extra cost. The Core lab only accepts pre-mixed fragment samples for fragment analysis. It does not affect the price if markers are multiplexed in each well; only the number of wells matters. The User enters sample names into the computer, otherwise the samples are labeled only by well number. Ask in the Core Lab for a sample sheet template if you want to enter names on your computer.

Real-Time PCR (Applied Biosystems 7900HT)

The user gets an introduction to the ABI 7900HT machine and software from a Core Lab Technician. Once this training is completed the user is free to sign-up on the calander and use the Real-Time PCR machine. Cost to use the 7900HT is $10/plate. The user provides plates, covers, chemistry etc. and sets up their own reactions. Click here for more information on the 7900HT system.

 

SpectraMax Spectrophotometer plates

Users can purchase 96-well UV Greiner plates at $6.25/each in the Core lab.
